    mixtures Environment Water quality Human health epidemiology Human health can be affected by exposure to a wide variety of chemicals Many processes that play a role in the onset of adverse health effects such as immunotoxicity endocrine disruption obesity and metabolic diseases carcinogenesis and reproductive toxicity can be assessed with CALUX reporter assays The sensitivity and high throughput compatibility of these assays makes them suitable both for mechanistic studies as well as screenings of large sample panels In many cases epidemiological studies and monitoring of human health effects are hampered by the limited amount of available sample material e g tissue blood In addition many analytical techniques are costly and time consuming to perform CALUX bioassays require small amount of material e g 1 ml human plasma for DR CALUX Especially in case of larger sample series CALUX analyses can be performed in a time and cost efficient manner due to the standardized high throughput compatible assay protocols Another important benefit of using CALUX assays is that they often give mechanistic insight in health effects that may otherwise only be correlated to a certain exposure from a statistical point of view Typical bioassays used for human monitoring and epidemiological studies

    information Directions International partners Login Print Food Feed Product Safety Human health epidemiology Chemicals pharmaceuticals Complex mixtures Environment Water quality Chemicals pharmaceuticals Correct safety evaluation of chemicals and pharmaceuticals is essential to protect consumers For industries realistic safety evaluation is of equal importance The use of experimental animals in safety evaluation is under pressure not only because of obvious ethical reasons but also because these tests are expensive time consuming low in throughput and often give unsatisfactory predictions of human toxicity Modern mechanistic bioassays provide insight in the modes of action of toxicants and can be used as alternative methods to assess chemical safety particularly when coupled to so called adverse outcome pathways AOP We have developed a wide range of such human cell based assays and we have automated them for the use in high throughput panels to assess safety of chemicals and pharmaceuticals These methods are particularly suited for use in integrated testing and read across procedures Other major applications include screening and bioactivity profiling to select new candidate drugs and chemicals with promising profiles with respect to safety Our latest developments focus on activity screens directed to beneficial effects including bioactivity profiling of herbal drugs and functional

    Publications Downloads Contact Contact information Directions International partners Login Print Food Feed Product Safety Human health epidemiology Chemicals pharmaceuticals Complex mixtures Environment Water quality Complex mixtures Biological effects by mixtures are typically hard to predict from chemical analytical data on the individual constituents which usually only cover part of the compounds actually present This can be circumvented by direct measurement of sum effects from the bioactive components in the mixture using CALUX reporter assays While most compounds are only tested if at all for their health effects in their pure form we are mostly exposed to complex mixtures of compounds Predicting health effects of mixtures is typically complicated for the interactions that may occur as well as the lack of information on individual components Effect based measurements using CALUX assays offer two important features for providing information on the effects of mixtures CALUX assays focus on effects instead of compounds This avoids bias towards targeted compounds CALUX assays directly measure the combined effects by the total mixture that is analyzed Results are therefore independent of prior toxicological knowledge on the individual constituents of a mixture and their mutual effects If desired a combination of chemical analytical and biology based techniques

    Complex mixtures Environment Water quality Water quality Being one of the key necessities of life water should be submitted to a thorough quality control Targeted chemical analysis of priority compounds can give an impression of the chemical quality but may not cover the entire load of pollutants that evoke biological effects CALUX assays are successfully applied in the identification of water samples that are polluted with biologically active toxicants even at low levels and address the complexity associated with the compound mixtures Current water quality monitoring practices mainly rely on targeted chemical analysis of selected set of compounds e g priority compounds This compound oriented approach however does not cover the entire load of pollution and may overlook the presence of biologically active chemicals Effect oriented testing approaches can overcome this limitation by measuring the effect of the total mixture of compounds that are present in a sample The CALUX assay panel is particularly suitable for this purpose since it detects a number of key effects in relation to water quality BDS provides routine services and participates various national and international projects in order to further implement a minimal panel of CALUX assays from a broad range of available assays
